Opioid antagonism in fentanyl antinociception experimental

چکیده

Among the most commonly used drugs to reduce pain and inflammation are nonsteroidal anti-inflammatory (NSAIDs) opioids. Opioids a wide group of including fentanyl fully synthetic opioid that is more potent morphine. The aim present study was evaluate antinociception in two murine tests, acetic acid writhing (WT) formalin hind paw (FHP). Likewise, involvement opioids antagonists: naltrexone, naltrindole nor-binaltorphimine induced activity estimated. antinociceptive evaluated from dose-response curves. intraperitoneal administration dose related effects with different potencies both tests. data revealed significant decrease analgesic effect by action WT FHP tests exception WT. antagonist reducing efficacy could possibly be other multiple mechanisms added inhibition activation MOR, DOR, KOR receptors. This demonstrates there functional interaction modulatory between assays paw. modulation seems mediated fentanyl. These results suggest than able effective models nociception inflammatory pain.

Sex differences in opioid antinociception.

Previous studies indicate that mu opioid agonists such as morphine may produce greater antinociception in male than in female rodents. The present study was designed to investigate the generality of this finding across dose, time and type of opioid agonist. In adult female and male Sprague-Dawley rats, time-effect curves were obtained for vehicle and three doses each of the mu agonists fentanyl...

Placenta ingestion by rats enhances y- and n-opioid antinociception, but suppresses A-opioid antinociception

Ingestion of placenta or amniotic fluid produces a dramatic enhancement of centrally mediated opioid antinociception in the rat. The present experiments investigated the role of each opioid receptor type (A, y, n) in the antinociception-modulating effects of Placental Opioid-Enhancing Factor (POEF—presumably the active substance). Antinociception was measured on a 52 jC hotplate in adult, femal...

Modification of local anesthetic-induced antinociception by fentanyl in rats.

In clinical practice, using the lowest doses of drugs for anesthesia or analgesia is the main goal. Opioid combinations with local anesthetics can be preferable for achieving adequate anesthesia or analgesia. The primary purpose of this study was to examine possible thermal antinociceptive effects of the opioid -fentanyl and the amide local anesthetics levobupivacaine and lidocaine when locally...

PoSSible invOIVement Of u1~OpiOid receptOrS in the fentanyl- Or mOrphine-induced antinOCiceptiOn at SupraSpinal and Spinal SiteS

Fentanyl has been shown to be a potent analgesic with a lower propensity to produce tolerance and physical dependence in the clinical setting. The present study was designed to investigate the mechanisms of fentanylor morphine-induced antinociception at both supraspinal and spinal sites. In the mouse tail-flick test, the antinociceptive effects induced by both fentanyl and morphine were blocked...
